探讨槲皮素对关节炎软骨细胞胆固醇代谢的影响及作用机制。方法 体外分离培养小鼠软骨细胞,分为对照组、模型组、低浓度组、高浓度组。CCK-8检测槲皮素对软骨细胞增殖的影响;胆固醇检测试剂盒检测细胞内总胆固醇(TC)水平;qRT-PCR检测骨关节炎软骨细胞中Cyp7b1、CH25H、RORα、Mmp3、Mmp13、Col2a1、Il-6的表达水平;Western blot检测软骨细胞中CYP7B1、CH25H、RORα、MMP3和MMP13的蛋白表达水平。结果 ①CCK-8结果显示浓度在10 μmol/L以下的槲皮素对软骨细胞没有毒性,而高浓度(25、50、100 μmol/L)的槲皮素会抑制软骨细胞增殖。②细胞内总胆固醇水平测定结果显示,使用槲皮素处理后的关节炎软骨细胞内总胆固醇水平较模型组低(P <0.05)。③qRT-PCR结果显示,槲皮素组中软骨细胞的Cyp7b1、CH25H、RORα、Mmp3、Mmp13、Il-6的表达水平较模型组降低,Col2a1的表达水平较模型组升高(P <0.05)。④Western blot结果显示药物干预组中CYP7B1、CH25H、RORα、MMP3、 MMP13的蛋白表达水平均比模型组降低(P <0.05)。结论 槲皮素可以通过CH25H/CYP7B1/RORα 信号通路调控关节炎软骨细胞中的胆固醇代谢,减轻关节炎软骨细胞中的炎症和细胞外基质降解。 |

Objective To investigate the mechanism of quercetin in cholesterol metabolism in osteoarthritic chondrocytes. Methods Mouse chondrocytes were isolated and cultured in vitro.The cells were divided into control group, model group, low concentration group, and high concentration groups. CCK-8 was performed to detect the effect of quercetin on the proliferation of chondrocytes. Total cholesterol test kit was used to detect cholesterol content in chondrocytes. qRT-PCR was performed to detect the expression of Cyp7b1, Ch25h, Rorα, Mmp3, Mmp13, Col2a1, and Il-6 in osteoarthritic chondrocytes. Western blotting was performed to detect the protein expression of CYP7B1, CH25H, RORα, MMP3, and MMP13 in chondrocytes. Results ①CCK-8 results showed that quercetin at concentrations below 10 μmol/L was not toxic to chondrocytes, while at high concentrations (25, 50, and 100 μmol/L) it inhibited chondrocyte proliferation. ②The results of intracellular total cholesterol content measurement showed that the total cholesterol content in the osteoarthritic chondrocytes after quercetin treatment was lower than that in the model group (P <0.05). ③qRT-PCR results showed that the expression levels of Cyp7b1, Ch25h, Rorα, Mmp3, Mmp13, and Il-6 in chondrocytes in the quercetin group were lower than those in the model group, and the expression level of Col2a1 was higher than that in the model group (P <0.05). ④Western blotting results showed that the protein expressions of CYP7B1, CH25H, RORα, MMP3, and MMP13 reduced in the drug intervention group compared to those in the model group (P<0.05). Conclusion Quercetin regulates cholesterol metabolism in osteoarthritic chondrocytes through the CH25H/CYP7B1/RORα signaling pathway and reduces inflammation and extracellular matrix degradation in osteoarthritic chondrocytes. |
