Objective To investigate the effects of Bing’s giant salamander ultra micro comminution on
bone metabolism, bone microstructure andWnt/β-catenin signaling pathway in ovariectomized rats. Methods75 SPF female SD rats aged 4-month-old were randomly divided into 5 groups: shamoperation group,
model group, ultra micro comminution group of Bing’s giant salamander. Osteoporosis, ordinary comminution group of Bing’s giant salamander, Osteoporosisand Xianlinggubao group. Except the sham operation
group, the animal model of osteoporosis was established by castration. After 12 weeks of drug
intervention, serum bone metabolism indexes were detected by ELISA, femoral bone microstructure
changes were measured by micro CT scanning, and the expression of Wnt/β-catenin signaling pathway
related proteins in bone tissue was measured by Western blot. Results After 12 weeks of drug
intervention, compared with the model group, ALP, OPG, PINP, BMD, BV/TV and in Notopterygii group and Xianlinggubao group TB.Th, Tb.N increased significantly, SMI and TB.SP and TRACP decreased
significantly, Wnt/β-Catenin The protein expression of catenin and Runx2 increased, but it was still lower
than that in sham operation group (P<0.05); The ultra micro comminution group was superior to the
ordinary comminution group in improving bone metabolism, bone microstructure and protein expression in
rats (P<0.05). Conclusion Bing’s giant salamander can delay the occurrence and development of
osteoporosis in ovariectomized rats, and ultra-fine powder is better than ordinary powder, and its
mechanism may be related to the activation of Wnt/β-Catenin signaling pathway. |
