Objective To explore the common genes between OP and AS by bioinformatics and to predict the traditional Chinese medicine (TCM) based on hub genes. Methods OP and AS related genes were selected from related disease databases (GeneCards, DISEASES, CTD). The common gene targets were obtained after de-intergration. Then, the PPI network of them was constructed through STRING database and Cytoscape software. CytoHubba plug-in was used to identify hub genes, which were used to predict TCM with preventive and curative effects using Coremine Medical database. Results There were 315 and 481 targets related to OP and AS, respectively, and 202 common targets were mapped. Results of enrichment analysis showed that the common mechanisms were mainly involved in immune and inflammatory response, PI3K-Akt pathway and MAPK pathway. Next, 10 hub common genes (including CD4, TLR2, TIMP1, CCL2, APOE, GAPDH, NFKBIA, IL4, CRP and TGFB1 )were obtained using CytoHubba, and a total of nine herbal drugs were predicted, including Scutellaria, ginseng, salvia miltiorrhiza and Panax notoginseng. Conslusion This study explored the potential co-pathogenesis of OP and AS at the molecular level. In addition, the TCM preventive and curative effects was preliminarily predicted, which could provide targets and research ideas for the development of TCM with concept of "simultaneous treatment of different diseases". |
