近十年来,间充质干细胞因其在骨组织工程、基因治疗和细胞治疗方面的巨大潜力而备受瞩目,其在基础研究和临床治疗应用的关键问题是细胞分化的控制,强调干细胞的状态和对其分化机制的调节对于细胞疗法在骨组织领域的发展至关重要。组蛋白乙酰化是表观遗传修饰中的一种调控方式,在不改变DNA序列的情况下调控基因的表达进而影响生物学表型,是干细胞成骨分化的决定性因素之一,也是迄今为止研究最为广泛的一种组蛋白修饰。与此同时,大量的研究表明组蛋白乙酰化水平与人类骨骼疾病密切相关。因此,本文综述了近十年来组蛋白乙酰化修饰酶及其调节剂对各来源的间充质干细胞成骨分化影响的研究进展,旨在阐明其潜在的调节机制,为临床上大量的骨代谢异常相关疾病提供进一步的治疗思路。 |

In the past decade, Mesenchymal stem cells (MSCs) have attracted much attention because of their great potential in bone tissue engineering, gene therapy and cell therapy in terms of the huge potential and high-profile, its application in basic research and clinical treatment is the key problem of the control of cell differentiation, the stressed state of stem cells and the differentiation mechanism of adjustment is vital for the development of cell therapy in the field of bone tissue. Histone acetylation is a regulatory mode of epigenetic modification, which regulates gene expression without changing DNA sequence and thus affects biological phenotype. It is one of the decisive factors of osteogenic differentiation of stem cells, and it is also the most widely studied histone modification so far. At the same time, numerous studies have shown that histone acetylation levels are closely related to human bone diseases. Therefore, this article reviews the research progress of histone acetylation modification enzymes and their modulators on osteogenic differentiation of mesenchymal stem cells from various sources in the past decade, aiming to clarify their potential regulatory mechanisms and provide further research ideas for bone regeneration. |
