      目的 研究温肾健脾、活血化瘀含药血清对骨髓间充质干细胞(BMSCs)成脂分化作用过程中生长激素(GH)/胰岛素样生长因子1(IGF-1)系统的调节机制。方法 将40只SPF级雄性SD大鼠用随机数字表法分成4组,依次为正常组、成脂诱导组、温肾健脾组、活血化瘀组。对大鼠进行7 d灌胃,制备含药血清,按一定比例将含药血清与完全培养基进行配比用以BMSCs成脂分化的培养。油红O染色观察BMSCs成脂分化情况后,用ELISA法测定脂肪细胞中GH、IGF-1的蛋白水平。结果 ①GH蛋白表达水平结果:与正常组比较,成脂诱导组低于正常组,差异不具有统计学意义(P>0.05)。与成脂诱导组相比,温肾健脾组、活血化瘀组结果均优于成脂诱导组,差异均具有统计学意义(P<0.05)。温肾健脾组与活血化瘀组相比,活血化瘀组结果优于温肾健脾组。②IGF-1蛋白表达水平结果:与正常组相比,成脂诱导组结果高于正常组,且差异具有统计学意义(P<0.05)。与成脂诱导组相比,温肾健脾组、活血化瘀组结果均低于成脂诱导组,且差异均具有统计学意义(P<0.05)。温肾健脾组、活血化瘀组相比,活血化瘀组结果高于温肾健脾组。结论 温肾健脾、活血化瘀中药可能通过GH/IGF-1系统发挥抑制BMSCs的成脂分化作用。

      Objective To study the regulatory mechanism of growth hormone (GH)/insulin like growth factor 1 (IGF1) system in the process of adipogenic differentiation of bone marrow mesenchymal stem cell cells (BMSCs) by serum containing drugs for warming the kidney and invigorating the spleen, promoting blood circulation, and removing blood stasis. Methods Forty SPF male Sprague-Dawley rats were randomly divided into four groups: normal group, lipid-inducing group, kidney-warming and spleen-strengthening group, and blood-activating and stasis-resolving group. The rats were fed for 7 days to prepare the serum containing drugs. The serum containing drugs was mixed with the complete culture medium in a certain proportion and used for the adipogenic differentiation of BMSCs. Adipogenic differentiation was observed with oil red O staining. The protein levels of GH and IGF-1 in adipocytes were determined with ELISA method. Results ①The expression levels of GH protein in the adipogenic induction group was significantly lower than that in the normal group (P>0.05). The results in the group of warming kidney and strengthening spleen and the group of promoting blood circulation and removing blood stasis were better than those in the lipid-induced group, with statistical significance (P<0.05). ②Results of IGF-1 protein expression in lipid induction group were higher than the normal group, and had statistical significance (P<0.05). The results in the group of warming kidney and strengthening spleen and the group of promoting blood circulation and removing blood stasis were lower than those in the lipid-induced group, and all of them had statistical significance (P<0.05). Conclusion Chinese herbs of warming kidney and strengthening spleen, promoting blood circulation and removing blood stasis may inhibit the adipogenic differentiation of BMSCs through GH/IGF-1 system.
