      铁是几乎所有生物体新陈代谢中必不可少的微量元素。但体内过量铁会产生细胞毒性,可使骨的强度和密度降低,铁过载可以通过抑制成骨细胞分化、增殖和刺激破骨细胞生成来破坏骨稳态。研究证实,铁螯合剂对铁过载导致的骨稳态失衡具有潜在的治疗价值。本文综述了铁过载对骨稳态的影响,阐明这一过程的细胞及分子机制,总结了铁螯合剂对成骨细胞及破骨细胞的不同影响,旨在为治疗铁过载造成的骨稳态失衡提供新的治疗靶点。
英文摘要:
      Iron is an essential trace element in the metabolism of almost all living organisms. However, excessive iron in the body can produce cytotoxicity, which can reduce the strength and density of the bone. Iron overload can disrupt bone homeostasis by inhibiting osteoblast differentiation, proliferation and stimulating osteoclast formation. It has been confirmed that iron chelating agents have potential therapeutic value for bone homeostasis imbalance caused by iron overload. In this paper, we review the effect of iron overload on bone homeostasis, elucidate the cellular and molecular mechanisms of this process, and summarize the different effects of iron chelating agent on osteoblasts and osteoclasts, aiming to provide a new therapeutic target for the treatment of bone homeostasis imbalance caused by iron overload.
