      目的 探讨香砂六君子汤防治绝经后骨质疏松症的主要机制。方法 采用随机数字表法对SD雌性大鼠进行随机分组,分为空白组、模型组、阳性对照组(仙灵骨葆胶囊组)及香砂六君子汤低、中、高剂量组。采用卵巢切除法(OVX)造模,造模成功后予以中药干预,八周后取材。通过Micro CT检测骨组织结构、生物力学仪检测股骨最大荷载力、ELISA法检测骨代谢、免疫组织化学法(IHC)检测骨形成相关蛋白及通路蛋白。结果 与模型组相比,各剂量组的香砂六君子汤均可恢复骨微观结构(P<0.05),尤以高剂量组明显,所以采用高剂量组作为本实验用药。其明显提高了大鼠股骨最大荷载力(P<0.05);显著上调了血清中ALP及PINP含量、下调了β-CTX含量水平(P<0.01);有效提高了骨小梁百分比(P<0.05);促进了调节骨形成的关键蛋白Runx 2表达和成骨通路相关蛋白Wnt及β-catenin表达(P<0.05)。结论 香砂六君子汤可恢复OVX大鼠股骨的骨小梁连续性及稳定性,促进骨形成,改善骨密度,进而防治PMOP,其作用机制可能与激活Wnt/β-catenin信号通路上调Runx2表达从而促成骨分化有关。

      Objective Research on the main mechanism of Xiangsha Liujunzi Decoction in preventing and treating postmenopausal osteoporosis. Methods SD female rats were randomly divided into 6 groups: blank group, model group, positive control group (Xian Ling Gu Bao capsule group), low, medium and high dose groups of Xiangsha Liujunji Decoction. Ovariectomy (OVX) method was used to make the model. After the model was successfully made, Chinese medicine was used to intervene. Materials were taken eight weeks later. MicroCT was used to detect bone tissue structure, biomechanical instrument was used to detect the maximum load of femur, ELISA was used to detect bone metabolism, and immunohistochemistry (IHC) was used to detect bone formation related proteins and pathway proteins. Results Compared with the model group, Xiangsha Liujunzi Decoction in each dose group could restore bone microstructure (P<0.05), especially in the high-dose group, so we used the high-dose group as the drug for this experiment; It also could significantly increase the maximum load on the femur of rats (P<0.05); Significantly increase and decrease the content of ALP and PINP in serum β-CTX content level (P<0.01); Effectively increase the percentage of bone trabeculae (P<0.05); Promote the expression of Runx 2, the key protein regulating bone formation, and the osteogenic pathway related protein Wnt and β- Catenin expression (P<0.05). Conclusion Xiangsha Liujunzi Decoction can restore the continuity and stability of the femoral trabecular bone of OVX rats, promote bone formation, improve bone density, and thus prevent and treat PMOP, and its mechanism may be related to the activation of Wnt/ β- Catenin signaling pathway up-regulates the expression of Runx2, which contributes to bone differentiation.
