      目的 探讨牵张应力调控Notch1促进骨髓间充质干细胞(BMSCs)增殖和成骨分化的机制。方法 制备并鉴定大鼠BMSCs。细胞分组设置为:0%拉伸幅度组、0%+DAPT组、5%拉伸幅度组、5%+DAPT组。运用CCK-8法测定BMSCs增殖;采用碱性磷酸酶、茜素红和油红O染色法检测细胞成骨和成脂分化;通过Western bloting检测Notch1和Jagged1蛋白表达。结果 5%拉伸幅度牵张应力可显著提高BMSCs增殖、碱性磷酸酶表达及钙化结节形成,抑制BMSCs成脂分化(P<0.05 或P<0.01),同时显著提高Notch1和Jagged1蛋白表达(P<0.01);DAPT可显著逆转牵张应力对BMSCs增殖、碱性磷酸酶表达、钙化结节形成、成脂分化及Notch1和Jagged1蛋白表达的影响(P<0.05 或P<0.01)。结论 5%拉伸幅度的牵张应力促进了BMSCs增殖和成骨分化,Notch1信号通路可能是牵张应力促进BMSCs增殖和成骨分化的靶点。

      Objective To investigate the mechanism of stretch stress regulating Notch1 to promote the proliferation and osteogenic differentiation of bone marrow mesenchymal stem cells (BMSCs). Methods Rat BMSCs were prepared and identified. The cells were divided into 0% stretch stress group, 0%+DAPT group, 5% stretch stress group, and 5%+DAPT group. Cell proliferation was determined by CCK-8 method. Alkaline phosphatase, alizarin red and oil red O staining were employed to detect the cell osteogenic and lipid differentiation. The protein expression of Notch1 and Jagged1 was detected by western bloting. Results The 5% stretch stress significantly increased the proliferation, the expression of alkaline phosphatase and the formation of calcified nodules, and inhibited the lipogenesis of BMSCs (P<0.05 or P<0.01). The expression of Notch1 and Jagged1 proteins was significantly increased by 5% stretch stress (P<0.01). DAPT could significantly reverse the effects of stretch stress on BMSCs proliferation, alkaline phosphatase expression, calcification nodule formation, lipid differentiation, and Notch1 and Jagged1 protein expressions (P<0.05 or P<0.01). Conclusion 5% stretch stress promotes the proliferation and osteogenic differentiation of BMSCs. Notch1 signaling pathway may be the target of stretch stress to promote the proliferation and osteogenic differentiation of BMSCs.
