      目的 探究龟板是否能够通过抑制炎症反应改善自然衰老小鼠的软骨终板退变。方法以3月龄小鼠作为年轻对照组,采用自然衰老模型,选取24月龄的小鼠12只,随机分为老年组与龟板组,每组6只。采用苏木素伊红染色及番红O-固绿染色实验明确自然衰老小鼠是否发生软骨终板退变;免疫组化检测IL-1β在各组小鼠软骨终板组织中的表达情况;免疫荧光染色检测Ⅱ型胶原蛋白在各组小鼠软骨终板组织中的表达情况。结果 与年轻对照组相比,老年组小鼠的HE染色表明其出现较为严重的软骨终板组织退变;与老年组相比,龟板组有一定程度的改善。番红O-固绿染色实验表明,与年轻对照组相比,老年组小鼠的软骨终板组织含水量减少;与老年组相比,龟板组小鼠的软骨终板组织胶原含量及含水量增加。组织免疫组化结果表明,老年组小鼠的软骨终板组织中IL-1β表达增加,药物干预后,龟板组小鼠的软骨终板组织中IL-1β表达减少。组织免疫荧光表明,与年轻对照组相比,老年组出现了明显的Ⅱ型胶原蛋白降解;药物干预后,龟板组有效地减少了Ⅱ型胶原蛋白的降解。结论 龟板可以通过抑制IL-1β减轻炎症,减少Ⅱ型胶原蛋白降解,从而改善自然衰老小鼠的软骨终板退变。

      Objective To investigate whether plastrum testudinis relieves cartilage endplate degeneration in senile mice by inhibiting inflammation reaction. Methods In this experiment, 3-month-old mice were used as a young control group. Using a natural aging model, 12 mice aged 24 months were selected and randomly divided into an aging group and a plastrum testudinis group, with 6 mice in each group. Hematoxylin eosin (H&E) staining and safranin O-fast green staining were used to determine whether the cartilage endplate degeneration occurred in senile mice. The expression of IL-1β in the cartilage endplate tissue of each group was detected with immunohistochemistry. The expression of type II collagen in the cartilage endplate tissue of each group was detected with immunofluorescence staining. Results Compared to that in the young control group, H&E staining of the senile mice showed more severe cartilage endplate tissue degeneration. Compared with the senile group, the plastrum testudinis group showed a certain extent of improvement. Safranin O-fast green staining showed that the water content of cartilage endplate tissue was reduced in senile group compared to that in the young control group. Collagen content and water content of cartilage endplate tissue increased in plastrum testudinis group compared to those in the senile group. Tissue immunohistochemistry results showed that IL-1β expression increased in the cartilage endplate tissues of mice in the senile group and decreased in the plastrum testudinis group after medical intervention. Tissue immunofluorescence results showed that significant type II collagen degradation occurred in senile group compared to that in the young control group. The degradation of type II collagen was effectively reduced after medicated intervention in the plastrum testudinis group. Conclusion Plastrum testudinis reduces inflammation and decreases type II collagen degradation by inhibiting IL-1β, thus relieving cartilage endplate degeneration in naturally aging mice.
