      骨骼是全身最大的器官之一,其发育及稳态维持异常可引起多种骨骼疾病,如骨骼遗传病、骨退行性疾病(如骨质疏松症、骨性关节炎)和骨折等。成纤维细胞生长因子受体(fibroblast growth factor receptors,FGFRs)家族重要成员FGFR1在骨骼发育和稳态维持中发挥关键作用。FGFR1功能增强点突变可引起囟门早闭综合征等骨骼遗传病,同时也参与调节骨形成、骨吸收过程以及关节炎的发生。此外,FGFR1在骨损伤修复不同阶段的骨骼相关细胞中均有表达,提示其参与骨修复过程。笔者总结了FGFR1在骨骼发育、稳态维持及损伤修复中的作用和机制研究进展,为从干预FGFR1信号角度治疗骨骼相关疾病提供借鉴。

      Bone is one of the largest organs in the body, and abnormal bone development and homeostasis can cause a variety of bone diseases, such as bone genetic diseases, degenerative diseases (such as osteoporosis, osteoarthritis) and fractures. FGFR1, a member of the FGFR family, plays important roles in skeleton development and homeostasis. Gain-of-function missense mutations in FGFR1 lead to bone genetic diseases such as craniosynostosis syndrome. FGFR1 also involved in the regulation of bone formation, bone resorption processes, and the development of osteoarthritis. In addition, FGFR1 was expressed in bone-related cells at different stages of bone injury repair, suggesting its involvement in the process of bone repair. This paper reviews the role and mechanism of FGFR1 in bone development, homeostasis maintenance and injury repair, providing new ideas for the treatment of bone diseases by interfering FGFR1 signaling.
