Objective To investigate the effect and mechanism of Uyghur medicine Qushi Jiangu Recipe on the prevention and treatment of postmenopausal osteoporosis based on calcium and phosphorus regulation. Methods Seventy SD rats were randomly divided into 7 groups: normal control group (Control group), sham operation group (Sham group), and surgical modeling groups including OVX group, positive control group (EVT group), low-dose group (QSJG-L group), medium-dose group (QSJG-M group), and high-dose group (QSJG-H group), with 10 rats in each group. OVX was performed in the surgical modeling groups. Rats received Bujiale (Estradiol Vital GmbH) 0.09 mg/(kg·d) in EVT group, 0.175 g/(kg·d) in QSJG-L group, 0.35 g/(kg·d) in QSJG-M group, and 0.7 g/(kg·d) in QSJG-H group, respectively, by gavage. Rats in other groups received the same amount of distilled water. The treatment lasted for 9 weeks. Micro-CT was used to detect bone mineral density. HE was used to observe pathological changes. ELISA was used to detect serum estrogen (E2), blood calcium (Ca), blood phosphorus (P), and other contents. Immunohistochemistry was used to detect renal 1α hydroxylase content. Results Compared to those in OVX group, bone mineral density (BMD) increased in QSJG-H group (P<0.01). Bone volume percentage, bone surface area, bone surface density, and trabecular number significantly increased. Bone surface area volume ratio and trabecular separation significantly decreased (P<0.01). HE results showed that trabecular interconnection was better, more complete, uniform thickness, and rare trabecular fracture traces in QSJG-H group. Serum E2, Ca, 1,25(OH)2D3 content increased (P<0.01). P and PTH contents significantly decreased (P<0.01). Renal 1α-hydroxylase expression significantly increased (P<0.01). Conclusion Uyghur medicine Qushi Jiangu recipe promotes the activation of renal 1α-hydroxylase to increase the level of 1,25 (OH) 2D3, inhibits PTH, and thus regulates calcium and phosphorus metabolism to play a role in preventing osteoporosis. |
