      N6-甲基腺嘌呤( N6-methyladenosine,m6A) RNA甲基化是生物体内最常见的信使RNA内部修饰,参与体内生理和病理过程。随着对m6A修饰的深入研究,其在多种疾病发病过程中的重要性备受关注。骨关节炎(osteoarthritis,OA)是一种关节软骨退行性疾病,近年来OA相关研究迅速增加,对其发病机制的探索是主要的研究方向之一。目前有关m6A修饰在OA发病机制的研究主要集中在保护ECM免受降解、抑制OA程序性细胞死亡、调节脂肪质量和肥胖相关蛋白、与非编码RNA互作影响和改善免疫微环境等五个方面。因此,本文将从这几方面重点阐释,以期为OA发病机制中m6A修饰的深入研究和基于表观遗传机制的骨科疾病诊疗思路及理论参考提供一定的依据。

      N6-methyladenosine (m6A) RNA methylation is the most common internal modification of messenger RNA in vivo and is involved in physiological and pathological processes in vivo. With the in-depth study of m6A modification,its importance in the pathogenesis of many diseases has been paid attention to. osteoarthritis (OA) is a degenerative disease of articular cartilage. Research on OA has increased rapidly in recent years,and the exploration of its pathogenesis is one of the main research directions. Current studies on m6A modification in the pathogenesis of OA mainly focus on five aspects:protection of ECM from degradation,inhibition of OA programmed cell death,regulation of fat mass and obesity-related proteins,interaction with non-coding RNA,and improvement of immune microenvironment. Therefore,this paper will focus on these aspects,in order to provide a certain basis for the in-depth study of m6A RNA methylation modification in the pathogenesis of OA and the treatment of orthopedic diseases based on epigenetic mechanism and theoretical reference.
