目的 探讨电针(electroacupuncture,EA)联合自体脂肪干细胞(adipose derived stem cells,ADSCs)移植对去卵巢大鼠骨质疏松的作用及对Wnt/β-catenin信号通路的影响。方法 将雌性SD大鼠随机分为假手术组、模型组、EA组、ADSCs组和EA+ADSCs组,每组10只。采用大鼠卵巢切除建立模型,在造模手术后2个月开始干预,EA组予双侧“脾俞”“肾俞”电针治疗,频率2 Hz,连续波,强度1 mA。每次10 min,每周3次,连续8周。ADSCs组将绿色荧光蛋白标记的ADSCs经尾静脉移植体内。每次1×106个,每周1次,连续8周。EA+ADSCs组予电针治疗,联合自体ADSCs移植,干预连续8周。检测大鼠股骨骨微结构及骨组织病理变化、血清骨代谢指标,股骨Wnt3a、β-catenin和Runx2基因及蛋白表达,及Runx2/β-catenin蛋白的共表达。结果 与假手术组比较,模型组大鼠骨密度显著降低(P<0.01),骨小梁间距变大;与模型组比较,EA组大鼠骨密度显著增高(P<0.01),骨小梁数量和厚度增加,Wnt3a、β-catenin、Runx2 mRNA和蛋白表达水平明显升高(P<0.01),Runx2/β-catenin蛋白表达显著升高(P<0.01);ADSCs组Runx2的mRNA和蛋白表达水平显著升高(P<0.01)。EA+ADSCs组骨小梁结构均优于ADSCs组和EA组,并且Runx2/β-catenin蛋白表达显著升高(P<0.01)。结论 ADSCs移植联合针灸可以有效改善卵巢摘除大鼠模型骨质疏松状况,这种作用可能和激活Wnt/β-catenin信号通路有关。 |

Objective To observethe effects of electroacupuncture combined with autologous adipose stem cells transplantation on osteoporosis and on the Wnt/β-catenin signaling pathway in ovariectomized rats. Methods Female SD rats were divided into sham operation group,model group,EA group,transplantation group and EA+ADSCs transplantation group,with 10 rats in each group. Two months after modeling,EA was applied to bilateral“Pishu”and“Shenshu”in the EA group, with oscillatory wave,2 Hz in frequency and 1mA in intensity. Each session lasted 10 minutes,3 times a week for 8 weeks. The ADSCs group transplanted green fluorescent protein-labeled ADSCs in vivo via the tail vein,1×106 each time,1 time per week for 8 weeks. The EA+ADSCs group was given electroacupuncture treatment,combined with ADSCs transplantation,for 8 weeks. Detection of morphological and structural changes in rat femur tissue,serum bone metabolism indexes,as well as detection of femur Wnt3a,β-catenin and Runx2 gene expression and proteins,and co-expression of Runx2/β-catenin proteins. Results Compared with the sham-operated group,rats in the model group had significantly lower BMD (P<0.01) and larger spacing of bone trabeculae;compared with the model group,rats in the EA group had significantly higher BMD (P<0.01),increased number and thickness of bone trabeculae,and significantly higher expression levels of Wnt3a,β-catenin,and Runx2 mRNAs and proteins(P<0.01),and significantly higher Runx2/β- catenin fluorescence intensity was significantly higher (P<0.01);the expression level of Runx2 was significantly higher in the ADSCs group (P<0.01).The bone trabecular structure of the EA + ADSCs group was superior to that of both the ADSCs group and the EA group,and the Runx2/β-catenin fluorescence intensity was significantly higher (P<0.01). Conclusion ADSCs transplantation combined with acupuncture can effectively improve osteoporosis in an ovariectomized rat model,and this effect may be related to the activation of the Wnt/β-catenin signaling pathway. |
