目的 通过检测BCL2及OPG/RANKL信号通路,探讨续苓健骨方抑制骨吸收的机制。方法 将3月龄SPF级雌性大鼠分成3组:假手术组(Sham组)、骨质疏松模型组(OVX组)、续苓健骨方组(XL组),制备骨质疏松模型。90 d后取材并检测左侧胫骨骨密度,HE和TRAP染色观察骨组织变化,ELISA检测血清IgG、IL-10、TGF-β、IL-6、IL-17水平,PCR和WB检测腰椎的BCL2、OPG和RANKL的基因表达。结果 OVX组较Sham组,骨形态学显示大鼠破骨细胞的生成数量增多、骨小梁数目减少、部分断裂、胫骨骨密度降低(P<0.01),IL-17和IL-6水平提高(P<0.01),IL-10、TGF-β水平降低(P<0.01),BCL2的表达降低(P<0.05),RANKL/OPG的比值升高(P<0.05);较OVX组,XL组骨小梁数目增加,结构明显改善,骨密度升高(P<0.05),IL-17、IL-6和IgG水平降低(P<0.05),IL-10、TGF-β水平增加(P<0.05),mRNA:BCL2表达升高(P>0.05),RANKL/OPG比值降低(P<0.01);蛋白BCL2表达升高(P<0.001),RANKL/OPG比值降低(P<0.05)。结论 续苓健骨方治疗OP的机制与其提高BCL2、降低RANKL/OPG的比值密切相关。 |

Objective To investigate the mechanism of inhibition of bone resorption by detecting BCL2 and OPG/RANKL signaling pathways. Methods Three-month-old female SPF rats were divided into three groups: sham operation group (Sham group), osteoporosis model group (OVX group), and Xuling strong-bone recipe group (XL group). The osteoporosis model was prepared. After 90 days, bone mineral density of the left tibia was measured. Bone tissue changes were observed with HE and TRAP staining. Serum levels of IgG, IL-10, TGF-β, IL-6, and IL-17 were detected with ELISA. The gene expressions of BCL2, OPG, and RANKL in the lumbar spine were detected with PCR and WB. Results Compared to that in the Sham group, bone morphology in the OVX group showed that the number of osteoclasts increased, the number of trabecular bones decreased, and some fractures were reduced, bone mineral density of the tibia decreased (P<0.01), the contents of IL-17 and IL-6 increased (P<0.01), the contents of IL-10 and TGF-β decreased (P<0.01), and the expression of BCL2 decreased (P<0.05). Compared to those in the OVX group, the ratio of OPG increased (P<0.05), the number of trabecular bones in the XL group increased, the structure was significantly improved, bone mineral density increased (P<0.05), the contents of IL-17, IL-6 and IgG decreased (P<0.05), the contents of IL-10 and TGF-β increased (P<0.05), the expression of mRNA:BCL2 increased (P>0.05), RANKL/OPG ratio decreased (P<0.01), the expression of BCL2 protein increased (P<0.001), and the RANKL/OPG ratio decreased (P<0.05). Conclusion The mechanism of Xuling strong-bone recipe in the treatment of OP is closely related to the increase of BCL2 and the decrease of RANKL/OPG ratio. |
