      骨重塑是一个维持骨骼系统稳态的连续过程,它取决于成骨细胞(osteoblast,OB)骨形成和破骨细胞(osteoclast,OC)骨吸收之间的稳态。骨吸收和骨形成的紧密耦合机制可能是由于在吸收过程中局部“耦合”因素的产生。TGF-β被认为是骨形成与先前骨吸收耦合的关键因素之一。TGF-β在抑制OC的形成和降低OC的活性方面发挥作用;同时,TGF-β也是非常有效的OB形成刺激剂,诱导OB的趋化、增殖和分化。然而,骨质疏松症(osteoporosis,OP)中TGF-β信号转导的详细机制仍未被完全阐明。因此,本文阐述了规范的TGF-β信号转导和非典型TGF-β信号转导机制,以及在骨细胞调控中的作用,期望为OP的治疗提供新的视角和方向。

      Bone remodeling is a continuous process to maintain homeostasis of the skeletal system, which depends on the homeostasis between osteoblast (OB) bone formation and osteoclast (OC) bone resorption. The tightly coupled mechanism of bone resorption and bone formation may be due to the production of local "coupling" factors during the resorption process. TGF-β is considered to be one of the key factors in coupling bone formation with previous bone resorption. TGF-β plays a role in inhibiting the formation and decreasing the activity of OC. TGF-β is also a very effective OB formation stimulator, inducing OB chemotaxis, proliferation and differentiation. However, the detailed mechanism of TGF-β signaling in osteoporosis (OP) has not been fully elucidated. Therefore, this paper describes the standardized TGF-β signal transduction and atypical TGF-β signal transduction mechanisms, as well as the role in bone cell regulation, hoping to provide a new perspective and direction for the treatment of OP.
