| Objective To explore the therapeutic effect and mechanism of Bushen Huoxue Recipe on diabetes osteoporosis (DOP). Methods By constructing a mouse model of type 1 diabetes, different doses of Bushen Huoxue prescription were given for 12 weeks , and then the fasting blood glucose value, weight change and other indicators of each group were studied to evaluate its effect. Micro CT scans were employed to examine the microstructure and histomorphometric features of the bone in the distal femur. HE staining enabled the detection of any pathological changes in the femur. Results Prescribing bushen huoxue had been shown to lower fasting blood sugar levels in diabetic mice (P<0.05), elevate their body weight (P<0.05), and alleviate diabetes symptoms.Significantly increased BV/TV, Tb.N and Tb.Th indexes(P<0.05)were observed in the trabecular structure, while a reduction of bone marrow cavity was noted; additionally, Tb.Sp and SMI values were significantly decreased(P<0.05). Elevate the levels of RUNX2 and COL1 expression in bone tissue, leading to an increase in the expression levels of RUNX2 and OPG protein in femoral tissue (P<0.05). Simultaneously, there was a significant decrease in serum inflammatory factors IL-6, IL-1β, and TNF-α (P<0.05), resulting in the alleviation of the inflammatory response in diabetic mice. Conclusion Bushen Huoxue prescription can prevent STZ-induced DOP, repair bone microstructure, and has a certain therapeutic effect on DOP. The mechanism may be to affect bone metabolism by activating the RUNX2/COL1 signaling pathway, thus playing a role in lowering blood glucose and alleviating osteoporosis. |
