| 骨质疏松症发病率高、危害大,已成为我国中老年人群的重要健康问题。随着研究的不断深入,SIRT3被发现在骨代谢中扮演重要角色,靶向SIRT3可能是治疗骨质疏松症的有效策略,例如激活具备抗氧化作用的SIRT3/SOD2轴。该文介绍了SIRT3的结构和功能并阐述了SIRT3促进成骨细胞生成、调控破骨细胞生成、促进骨髓间充质干细胞分化和抑制其衰老的作用与机制,以及目前靶向SIRT3影响骨代谢或骨质疏松症的研究进展,以期能为骨质疏松症的发生机制和治疗靶点提供新的见解。 |

| Osteoporosis is highly prevalent and harmful, and has become an important health problem in the middle-aged and elderly population in China. With the deepening of research, SIRT3 has been found to play an important role in bone metabolism. Targeting SIRT3 may be an effective strategy for the treatment of osteoporosis. For example, activating the SIRT3/SOD2 axis may possess antioxidant effects. This article introduces the structure and function of SIRT3 and describes the roles and mechanisms by which SIRT3 promotes osteoblastogenesis, regulates osteoclastogenesis, promotes bone marrow mesenchymal stem cell differentiation, and inhibits their senescence. Also, the current progress of research in targeting SIRT3 to affect bone metabolism or osteoporosis is reviewed, aiming to provide new insights in the mechanisms and therapeutic targets in the development of osteoporosis. |
