      骨组织重建过程中,成骨与成血管密切耦合,共同维系骨的生理代谢和平衡。低氧诱导因子-1α(Hypoxia-Inducible Factor 1-alpha,HIF-1α)及血管内皮生长因子(Vascular Endothelial Growth Factor,VEGF)共同构成的信号轴,是骨形成及血管新生过程中的核心调控枢纽。HIF-1α通过上调VEGF表达,引导新生血管向成骨区生长,从而为骨修复提供氧气、营养及成骨细胞来源,实现骨–血管的同步重塑。近年来,大量研究证实,苷类化合物可通过调控HIF-1α/VEGF轴改善骨微环境,促进血管与骨的同步重塑,在骨–血管耦联的调控中展现出显著潜力。本文综述了苷类化合物调控HIF-1α/VEGF轴介导的成骨–成血管耦联机制及相关研究进展,旨在为骨再生及中药创新应用提供理论依据。
英文摘要:
      During bone tissue remodeling, osteogenesis and angiogenesis are closely coupled, jointly maintaining bone metabolism and homeostasis. The signaling axis composed of hypoxia-inducible factor-1α (HIF-1α) and vascular endothelial growth factor (VEGF) serves as a central regulatory hub in both bone formation and angiogenesis. HIF-1α promotes VEGF expression, guiding neovascularization toward osteogenic sites, thereby supplying oxygen, nutrients, and osteoprogenitor cells for bone repair and achieving synchronous bone–vascular remodeling. In recent years, numerous studies have demonstrated that glycosides can regulate the HIF-1α/VEGF axis to improve the bone microenvironment, enhance coupled angiogenesis and osteogenesis, and show remarkable potential in the regulation of bone–vascular coupling. This review summarizes the mechanisms and research progress of glycosides in regulating HIF-1α/VEGF axis-mediated osteogenesis–angiogenesis coupling, aiming to provide a theoretical basis for bone regeneration and innovative applications of traditional Chinese medicine.
