| 目的 研究补肾化痰方改善肥胖型多囊卵巢综合征(polycystic ovary syndrome,PCOS)小鼠骨骼肌生物学功能的效应与潜在机制。方法 50只雌性2月龄小鼠随机分组,PCOS组小鼠于背部皮下植入来曲唑缓释片,对照组(Con)小鼠植入空载缓释片。各组小鼠均给予高脂饲料饲养5周,同时给予低、高剂量补肾化痰方干预5周。实验结束后,检测空腹血糖、口服糖耐量试验(OGTT)等;评价卵巢、骨骼肌组织学病理;测量抓力、骨骼肌湿重系数;测定小鼠血清硫酸脱氢表雄酮DHEA-S及胰岛素含量。结果 与Con组相比,PCOS组小鼠体质量(P<0.01)、空腹血糖(P<0.05)、血清胰岛素水平(P<0.05)均显著性增高,OGTT后血糖显著升高(P<0.01),卵巢发生囊性扩张等典型改变。补肾化痰方干预后,空腹血糖、OGTT后血糖水平以及血清胰岛素均显著下降,卵巢无明显闭锁囊性卵泡,小鼠抓力、骨骼肌湿重系数、骨骼肌肌纤维面积增加,骨骼肌组织AKT(Ser473)位点磷酸化水平显著性上调。结论 补肾化痰方能够改善肥胖型PCOS小鼠胰岛素抵抗与骨骼肌功能,提示其对于临床PCOS患者可能有额外的骨骼肌获益。 |

| Objective To investigate the effects and potential mechanisms by which the Bushen Huatan formula (BHF) improves skeletal muscle biological function in obese polycystic ovary syndrome (PCOS) mice. Methods Fifty female mice (2 months old) were randomly allocated into groups. Mice in the PCOS group were subcutaneously implanted with letrozole slow-release pellets, whereas those in the control group (Con) received empty pellets. All groups were fed a high-fat diet for 5 weeks and simultaneously treated with low or high doses of BHF for 5 weeks. At the end of the experiment, fasting blood glucose and glucose tolerance were measured. Ovarian and skeletal muscle histopathological changes were examined. Grip strength and skeletal muscle wet-weight ratio were determined. Serum levels of dehydroepiandrosterone sulfate (DHEA-S) and insulin in the mice were also measured. Results Compared with the Con group, body weight in the PCOS group increased significantly (P<0.01), as did fasting blood glucose (P<0.05), and serum insulin levels (P<0.05). Blood glucose levels significantly increased after the OGTT (P<0.01). Ovaries displayed cystic dilation and other typical alterations. After treatment with BHF, fasting blood glucose, blood glucose levels after OGTT, and serum insulin levels were significantly reduced, and no obvious atretic cystic follicles were observed in the ovaries. Additionally, in grip strength, skeletal muscle wet-weight ratio, and muscle fiber cross-sectional area were increased in BHF-treated mice. The phosphorylation level of AKT(Ser473) in skeletal muscle tissue was significantly upregulated. Conclusion Bushen Huatan formula can ameliorate insulin resistance and improve skeletal muscle function in obese PCOS mice, suggesting it may offer additional skeletal muscle benefits for clinical PCOS patients. |
