      骨质疏松症(osteoporosis,OP)对个人健康和医疗卫生领域构成极大挑战,是国内外骨科学领域研究的重点内容。PI3K/Akt通路通过影响骨稳态,即参与骨吸收和骨形成之间的平衡,与OP的进展密切相关。根据现有研究,PI3K/Akt通路主要通过mTOR、GSK-3β、FOXO1 3个下游效应分子,MEK/ERK(MAPK)、NF-κB和Wnt/β-catenin信号串扰参与OP的进展。通过归纳下游效应分子和信号串扰在成骨细胞(osteoblast,OB)、骨髓间充质干细胞(bone marrow mesenchymal stem cell,BMSC)、破骨细胞(osteoclast,OC)等靶点的作用机制和影响,本文为OP中研究机制尚少的FOXO1和PI3K/Akt-MAPK串扰、靶点尚未明确的PI3K/Akt-Wnt/β-catenin串扰提供理论参考。
英文摘要:
      Osteoporosis (OP) poses a great challenge to personal health and the field of medical and health care. It is a key research content in the field of orthopedics both at home and abroad. PI3K/Akt pathway is closely related to the progression of OP by affecting bone homeostasis, i.e, participating in the balance between bone resorption and bone formation. According to the existing research, PI3K/Akt pathway is mainly involved in the progression of OP through three downstream effector molecules, namely mTOR, GSK-3β and FOXO1, as well as the signal crosstalk of MEK/ERK (MAPK), NF-κB, and Wnt/β-catenin. By summarizing the action mechanisms and influences of the downstream effector molecules and signal crosstalk on targets such as osteoblasts (OB), bone marrow mesenchymal stem cells (BMSC), and osteoclasts (OC), this paper provides a theoretical reference for the FOXO1 and PI3K/Akt-MAPK crosstalk with relatively few research mechanisms in OP, as well as the PI3K/Akt-Wnt/β-catenin crosstalk whose targets have not been clearly defined.
