| 随着对肝骨串扰的深入研究,人们愈加关注针对不同类型骨质疏松症(osteoporosis, OP)的靶向干预。研究表明,肝损伤导致的OP揭示了肝骨之间新的调节机制。尽管“肝-骨轴”在原发性和继发性OP中的作用尚未完全明确,但探索其调控机制对于开发治疗肝脏和骨骼相关疾病的新方法至关重要。本文综述了多种内分泌因子(如IGF-1、FGF21、铁调素等)和关键基因(如SIRT2、TFR2、SREBP2等)在“肝-骨轴”调节中的潜在作用。除了炎症的经典代谢途径外,“肝-骨轴”在铁代谢、胆固醇代谢、脂质代谢和免疫代谢中的作用也需要进一步研究,以阐明其在OP中的具体调节机制。肝脏代谢异常对骨骼代谢失衡的影响强调了肝脏代谢稳态在OP发病机制中的重要性,为预防、诊断和治疗肝脏和骨骼相关疾病提供了新的方向。针对现有“肝-骨轴”机制,中西医从不同层面治疗OP,通过对“肝-骨轴”的进一步探索,有望为OP患者提供全面和创新的治疗方案。 |

| With the in-depth study of hepato-bone crosstalk, more and more attention has been paid to targeted interventions for different types of osteoporosis (OP). Studies have shown that OP due to liver injury reveals a new regulatory mechanism between liver and bone. Although the role of the hepatooskeletal axis in primary and secondary OP is not fully understood, exploring its regulatory mechanisms is essential for the development of new approaches for the treatment of liver and bone-related diseases. This article reviews the potential roles of various endocrine factors (e.g., IGF-1, FGF21, hepcidin, etc.) and key genes (e.g., SIRT2, TFR2, SREBP2, etc.) in hepato-bone axis regulation. In addition to the classical metabolic pathways of inflammation, the role of the hepato-bone axis in iron metabolism, cholesterol metabolism, lipid metabolism, and immune metabolism also needs to be further studied to elucidate its specific regulatory mechanisms in OP. The effect of abnormal liver metabolism on bone metabolic imbalance emphasizes the importance of hepatic metabolic homeostasis in the pathogenesis of OP, and provides a new direction for the prevention, diagnosis and treatment of liver and bone-related diseases. In view of the existing hepato-bone axis mechanism, TCM and Western medicine treat OP from different levels. Future possibilities may involve drugs that target communication between hepatocytes and bone cells, as well as modulating the activity of key molecules and enzymes. Through further exploration of the hepato-bone axis, it is expected to provide more comprehensive and innovative treatment options for patients with OP. |
