      目的 探讨T2DM绝经后女性LRP5rs41494349、rs2306862位点基因多态性及突变与骨代谢的关系。方法 收集新疆绝经后女性根据血糖及BMD分为4组:糖耐量正常伴骨量正常组(A组),T2DM伴骨量正常组(B组),糖耐量正常伴骨量异常组(C组),T2DM伴骨量异常组(D组)。检测各临床指标,测定LRP5rs41494349、rs2306862位点基因型。结果 1、与A组相比:B组、D组FPG、HbA1c%升高,C组、D组TG、BMD(L1-4)、BMD(股骨颈)降低(P<0.05)。2、rs2306862位点,突变型(CT/TT)与野生型(CC)相比,B组P升高,D组BMD(L1-4)、P降低(P<0.05);rs41494349位点,突变型(AG/GG)与野生型(AA)相比,D组的BMD(L1-4)降低(P<0.05)。3、多元线性回归分析显示:绝经年限、BMI是BMD(L1-4)及BMD(股骨颈)的影响因素,TG是BMD(L1-4)的影响因素,两位点的多态性是BMD(股骨颈)的影响因素。结论 LRP5rs41494339、rs2306862位点基因多态性及突变通过影响BMD及骨代谢指标,可能参与了绝经后女性的T2DM合并OP的发生和发展。

      Objective: To investigate the relationship between polymorphism and mutation of LRP5rs41494349,rs2306862 locus gene and bone mineral density (BMD) in postmenopausal women with type 2 diabetes (T2DM).Methods:The postmenopausal women in Xinjiang were divided into 4 groups :Normal glucose tolerance with normal bone mass group (group A), T2DM with normal bone mass group (group B), normal glucose tolerance with abnormal bone mass group (group C), T2DM with abnormal bone mass group (group D).The clinical indexes and LRP5rs41494349,rs2306862 locus gene polymorphism.were measured. Data was calculated by SPSS19.0 statistical software.Results: 1.Compared with group A:the leve of FPG and HbA1c% in group B and D were increased.The leve of TG,BMD(L1-4) and BMD (femoral neck) in group C and D were reduced.2. Mutant (CT/TT) compared with wild-type (CC) in rs2306862 locus:P was increased in group B,BMD (L1-4)and P were decreased in group D(P <0.05);Mutant (AG/GG) compared with wild-type (AA) in rs41494349 locus:BMD (L1-4) in group D was decreased.3.Multivariate linear regression analysis:menopause years and BMI were the influencing factors of BMD (L1-4) and BMD (femoral neck).TG was the influencing factors of BMD (femoral neck). Rs41494339 and rs2306862 locus in LRP5 gene polymorphism were the influencing factors of BMD (femoral neck).Conclusion: LRP5rs41494339, rs2306862 locus gene polymorphisms and mutations may be involved in the occurrence and development of T2DM with OP by affecting BMD and bone metabolism markers in postmenopausal women.
