| 目的：探究外敷温经药酒联合针刺对骨质疏松大鼠骨钙素、成骨细胞活性及Notch信号通路的影响。方法：选取SPF级雌性大鼠40只,分为健康组、模型组、针刺组、联合组,平均10只/组。除健康组外其余均建立骨质疏松症(OP)模型。建模后健康组、模型组不予处理,针刺组采用针刺治疗,5 d/周,1次/d,20 min/次,共12周。联合组在针刺治疗基础上外敷温经药酒,2次/d,连续治疗12周。采用ELISA法检测骨钙素(OC) 、吡啶酚(PYD)、Ⅰ型前胶原羧基端肽(PICP),采用计算机断层扫描检测骨体积结构,TUNEL法检测成骨细胞凋亡, HE染色观察病理改变,Western blot法检测Notch1、Notch2、Jagged1、Jagged2蛋白表达。结果：与健康组比较,模型组PYD、Tb.Sp、成骨细胞凋亡率升高,OC、PICP、Tb.Th、Notch1、Notch2、Jagged1、Jagged2降低(P＜0.05)；与模型组比较,针刺组PYD、Tb.Sp、成骨细胞凋亡率降低,OC、PICP、Tb.Th、Notch1、Notch2、Jagged1、Jagged2升高(P＜0.05)；与针刺组比较,联合组PYD、Tb.Sp、成骨细胞凋亡率降低,OC、PICP、Tb.Th、Notch1、Notch2、Jagged1、Jagged2升高(P＜0.05)；健康组大鼠骨形态结构良好；模型组骨小梁显著降低且具有排列紊乱、断裂现象；针刺组骨形态结构有所改善,但排列仍不规则并伴随部分断裂；联合组骨形态结构排列趋于规则,连续性与完整性改善良好。结论：外敷温经药酒联合针刺对骨质疏松大鼠的骨钙素、成骨细胞活性及Notch信号通路等的改善作用良好,建议推广使用。 |

| : Objective: To investigate the effects of external application of warm meridian medicinal wine combined with acupuncture on osteocalcin, osteoblast activity, and Notch signaling pathway in osteoporosis rats.Method: Forty SPF grade female rats were selected and divided into healthy group, model group, acupuncture group, and combination group, with an average of 10 rats per group. Except for the healthy group, all others established OP models. After modeling, the health group and model group were not treated. The acupuncture group received acupuncture treatment for 5 days/week, once a day, for 20 minutes/time, for a total of 12 weeks. On the basis of acupuncture treatment, the combined group applied warm meridian medicinal wine twice a day for 12 consecutive weeks. Using ELISA method to detect osteocalcin (OC), pyridine phenol (PYD), and type I procollagen carboxyl terminal peptide (PICP), using computer tomography to detect bone volume structure, TUNEL method to detect osteoblast apoptosis, HE staining to observe pathological changes, and Western blot method to detect protein expression of Notch1, Notch2, Jagged1, and Jagged2.Result: Compared with the healthy group, the model group showed an increase in PYD, Tb. SP, and osteoblast apoptosis rate, while the OC, PICP, Tb. Th, Notch1, Notch2, Jagged1, and Jagged2 proteins decreased (P<0.05); Compared with the model group, the acupuncture group showed a decrease in PYD, Tb. SP, and osteoblast apoptosis rate, while OC, PICP, Tb. Th, Notch1, Notch2, Jagged1, and Jagged2 proteins increased (P<0.05); Compared with the acupuncture group, the combined group showed a decrease in PYD, Tb. SP, and osteoblast apoptosis rate, while OC, PICP, Tb. Th, Notch1, Notch2, Jagged1, and Jagged2 proteins increased (P<0.05); The bone morphology and structure of the healthy group rats were good; The bone trabeculae in the model group significantly decreased and exhibited disordered arrangement and fracture phenomena; The bone morphology and structure of the acupuncture group improved, but the arrangement was still irregular and accompanied by partial fracture; The arrangement of bone morphology and structure in the combined group tends to be regular, with good improvement in continuity and integrity.Conclusion: The combination of external application of Wen Jing Yao Jiu and acupuncture has a good improvement effect on osteocalcin, osteoblast activity, and Notch signaling pathway in osteoporosis rats, and it is recommended to promote its use. |
