| 目的 考察阿法骨化醇对糖尿病小鼠骨组织的作用。方法 使用腹腔注射链脲佐菌素诱导糖尿病模型。ELISA方法检测血清骨代谢指标。微焦点定量CT检测骨密度、并经三维重建获得微观结构。使用RT-PCR、蛋白印迹分别检测基因、蛋白表达。结果 糖尿病小鼠血清B-ALP,TRACP-5b、尿钙水平升高，股骨远端的骨密度降低, 骨组织RAS组分AGT , Renin-R ,ANG Ⅱ表达显著上调。阿法骨化醇降低糖尿病小鼠血清TRACP-5b、尿钙水平，提高骨密度，抑制骨组织高RAS活性，显著下调破骨因子CAⅡ , MMP-9表达。结论 阿法骨化醇通过抑制糖尿病小鼠骨组织RAS活性而防治糖尿病性骨质疏松症。 |

| Objective To investigate the effect of alfa-calcidol on bone tissue in diabetic mice.Methods The diabetic mouse model was established using intrapentoneal injection of streptozotocin .Serum bone metabolic markers were detected using ELISA .Bone mineral density was detected using micro-CT, and the three-dimensional microstructure was reconstructed .Gene expression was detected using RT-PCR, and protein expression was detected using Western blotting.Results The serum levels of B-ALP and TRACP-5b and the urinary level of calcium increased in diabetic mice .The bone mineral density of the distal femur reduced，while the expression of RAS components，including AGT，Renin-R, and ANG II，increased significantly. Alfa-calcidol could reduce the level of serum TRACP-5b and urine Ca，improve the bone mineral density，inhibit the high activity of renin-angiotensin system(RAS ) in bone tissue，and down-regulate the expression of osteoclastic factors，including CAII and MMP-9，in diabetic mica. Conclusion Alfa-calcidol can prevent diabetes induced osteoporosis by inhibiting the RAS activity in hone tissue in diabetic mice. |
