      目的 探讨GLP-1受体激动剂艾塞那肽(Exenatide)对人成骨肉瘤MG-63细胞株增殖及成骨分化的作用。方法 将 MG-63细胞制成单细胞悬液,接种于细胞培养板,分别用不同浓度的艾塞那肽(0 mol/L、10-9 mol/L、10-8 mol/L、10-7 mol/L)处理细胞,24 h后采用CCK-8比色法检测MG-63细胞增殖率,并将艾塞那肽处理后的细胞裂解并取上清,使用碱性磷酸酶测定试剂盒测定AKP活力。结果(1)艾塞那肽可促进MG-63细胞增殖(P<0.05),但随着GLP-1受体激动剂浓度升高,细胞增殖程度降低(P <0. 05),低浓度的艾塞那肽(10-9 mol/L)促增殖能力最强。(2)艾塞那肽不同浓度处理后各组AKP活力增加(P<0.05),而且低浓度的艾塞那肽(10-9 mol/L)升高AKP活力最明显(P<0.05)。结论 艾塞那肽能促进人成骨肉瘤MG-63细胞的增殖及成骨分化,且低浓度的艾塞那肽(10-9mol/L)促增殖分化能力最强。

      Objective To investigate the effect of GLP-1 receptor agonist, exenatide, on the proliferation and osteoblastic differentiation by the human osteosarcoma MG63 cells. Methods Different concentrations of exenatide (0 mol/L, 10-9 mol/L, 10-8 mol/L, and 10-7 mol/L, respectively) were additioned to MG-63 cells cultured in vitro. Cell proliferation was measured with cell counting kit-8 (CCK-8) after 24 h. Moreover, the supernatants of exenatide-treated MG-63 cells were collected. AKP activity was tested using AKP kit. Results (1) Exenatide promoted proliferation of MG-63 cells (P< 0. 05) .With the increase of the concentration, the proliferation rate of MG-63 cells declined (P < 0.05 ).The most stimulation effect was achieved at the concentration of 10-9mol/L. (2) The AKP activity of MG-63 cells increased after treatment with exenatide, and the effect was the most obvious at the lower the concentration of exenatide ( P < 0.05 ). Conclusion Exenatide promotes proliferation and osteoblastic differentiation by human osteosarcoma MG-63 cells. Exenatide at the lower concentration (10-9 mol/L) has the strongest effect on promoting the proliferation and differentiation.
