      目的 探讨槲皮素对类风湿性关节炎成纤维样滑膜细胞(MH7A)炎症因子和基质金属蛋白酶的表达的影响及可能机制。方法 选用人类风湿性关节炎成纤维样滑膜细胞(MH7A)为研究对象,TNF-α诱导MH7A细胞,用不同浓度的槲皮素干预,RT-PCR法检测对IL-1β、IL-6、IL-8、MMP-1、MMP-3、MMP-13的变化的影响,Western blot法检测对NF-κB的影响。结果 槲皮素浓度依赖性地降低TNF-α诱导的MH7A细胞IL-1β、IL-6、IL-8、MMP-1、MMP-3、MMP-13的表达,对TNF-α诱导的MH7A细胞NF-κB的磷酸化有明显的抑制作用。结论 槲皮素可有效地减轻类风湿性关节炎成纤维样滑膜细胞炎症因子及基质金属蛋白酶的表达,其机制可能和抑制NF-κB的表达相关,槲皮素是治疗类风湿性关节炎的潜在有效药物。

      Objective To investigate the effect of quercetin on the expression of inflammatory factors and matrix metalloproteinases in rheumatoid arthritis fibroblast-like synoviocytes and its possible mechanism. Methods Human rheumatoid arthritis synovial fibroblasts (MH7A) were the research object, and MH7A were induced by TNF- alpha, with different concentrations of quercetin. RT-PCR method was used to detect the changes of IL-1, IL-6, IL-8, MMP-1, MMP-3 and MMP-13. NF-kappa B signaling was detected by Western blot. Results Quercetin concentration dependently reduced the expression of IL-1 beta, IL-6, IL-8, MMP-1, MMP-3 and MMP-13 in MH7A cells induced by TNF- alpha, and significantly inhibited the phosphorylation of NF- kappa B induced by TNF- alpha in MH7A cells. Conclusion Quercetin can effectively reduce the expression of inflammatory factors and matrix metalloproteinases in fibroblast-like synoviocytes of rheumatoid arthritis, and its mechanism may be related to the inhibition of NF-κB expression. Quercetin is a potentially effective drug for the treatment of rheumatoid arthritis.
